چکیده

Waterand alkali-soluble hemicelluloses were isolated with hot water and 10% KOH at 25 °C from dewaxed and delignified Salix psammophila, respectively. The alkali-soluble hemicelluloses were then successively subfractionated by neutralization and subjected to gradual precipitation in the end, using ethanol concentrations of 15, 30, 45, 60, 75 and 90%, respectively. Chemical composition, physico-chemical properties and structures of the 8 precipitated hemicellulosic fractions obtained were established by sugar analysis, molecular weight determination, FT-IR, H, C and 2D HSQC NMR. It was found out that the water-soluble hemicelluloses were more branched, having a low molecular weight (6060 g mol), whereas the alkali-soluble hemicelluloses were less branched, having higher molecular weights (17110-85540 g mol), with xylose and uronic acid as the main sugar components. In addition, the less branched hemicelluloses with large molecules were precipitated in lower ethanol percentages while, with increasing ethanol concentration, more branched hemicelluloses with low molecular weights were obtained. That is why, linear hemicelluloses could be recovered at lower ethanol concentrations, and more branched hemicelluloses could be obtained at higher ethanol concentrations. According to FT-IR, H, C and 2D HSQC NMR studies, the alkali-soluble hemicelluloses had a main structure composed of a (1→4)-linked β-D-xylopyranosyl backbone with 4-O-methyl-α-D-glucuronic acid attached to the O-2 of the xylose residues.

Seasonal Dynamics of Water Use Strategy of Two Salix Shrubs in Alpine Sandy Land, Tibetan Plateau

Water is a limiting factor for plant growth and vegetation dynamics in alpine sandy land of the Tibetan Plateau, especially with the increasing frequency of extreme precipitation events and drought caused by climate change. Therefore, a relatively stable water source from either deeper soil profiles or ground water is necessary for plant growth. Understanding the water use strategy of dominant ...

De novo transcriptome assembly, development of EST-SSR markers and population genetic analyses for the desert biomass willow, Salix psammophila

Salix psammophila, a sandy shrub known as desert willow, is regarded as a potential biomass feedstock and plays an important role in maintaining local ecosystems. However, a lack of genomic data and efficient molecular markers limit the study of its population evolution and genetic breeding. In this study, chromosome counts, flow cytometry and SSR analyses indicated that S. psammophila is tetra...

Selection of Reliable Reference Genes for Gene Expression Analysis under Abiotic Stresses in the Desert Biomass Willow, Salix psammophila

Salix psammophila is a desert shrub willow that has extraordinary adaptation to abiotic stresses and plays an important role in maintaining local ecosystems. Moreover, S. psammophila is regarded as a promising biomass feedstock because of its high biomass yields and short rotation coppice cycle. However, few suitable reference genes (RGs) for quantitative real-time polymerase chain reaction (qR...

Secondary metabolites as stimulants and antifeedants of Salix integra for the leaf beetle Plagiodera versicolora.

Plagiodera versicolora, a willow beetle living on S. sachalinensis, is found on S. integra during early June in Hokkaido Island, Japan. This insect selects several species of willows (Salix), including S. integra as host plant in Honshu Island of Japan. To determine the reasons for the limited distribution of this beetle on the willows of Hokkaido, the feeding preference of the insect to leaves...
